Relationship between Interleukin (IL)-6 Gene (AT) Polymorphism, Serum Il-6 Levels and Bone Mineral Density in Postmenopausal Korean Women

Abstract
Objectives: To investigate the relationship between a polymorphic AT rich minisatelite repeat in IL-6 gene, and bone mineral density (BMD) in postmenopausal Korean women.

Methods: A variable number tandem repeat (VNTR) in the IL-6 gene was examined by polymerase chain reaction (PCR) and electrophoresis in 274 postmenopausal Korean women. Serum IL-6, bone alkaline phosphatase (BAP) and CrossLaps (CTX) were measured by enzyme-linked sorbent assay and immunoassay, and BMD at the lumbar spine and proximal femur by dual energy X-ray absorptiometry.

Results: Four alleles in IL-6 gene were identified; B1 (760 bp), B2 (680 bp), B3 (640 bp), and B4 (610 bp). Five genotypes were found and B3 homozygote was the most frequent type. No differences in serum levels of IL-6, BAP, CTX and BMD at the lumbar spine, femoral neck, Ward¡¯s triangle and trochanter were observed between B3 homozygotes and B3 heterozygotes. There were no differences in the distribution of IL-6 B3 genotypes among normal, osteopenic, and osteoporotic postmenopausal women.

Conclusion: A IL-6 AT rich minisatelite is not associated with BMD in Korean women.
